Spermidine, honestly — promising cohort, null trial
Spermidine is a fashionable longevity supplement built on genuinely intriguing data — and one telling null trial. The honest headline: people who eat more spermidine-rich food live longer in observational studies, but the one human supplement trial found no benefit, and diet likely explains a lot.
From autophagy to the supplement aisle
Spermidine is a compound found in foods like wheat germ, legumes, mushrooms and aged cheese. It can trigger autophagy — the cell’s recycling of damaged parts — which is a plausible anti-ageing mechanism, and that story drove its popularity as a supplement.
The human evidence, though, splits sharply between promising observation and a disappointing trial.
What the evidence shows
Association strong, causation unproven.
- Diets high in spermidine track with lower mortality — In a well-known cohort, higher dietary spermidine intake was associated with lower all-cause mortality — the top intake group looked years "younger" in risk. Striking, but observational. (emerging)
- The human supplement trial was null — A 12-month randomised trial of wheat-germ spermidine in older adults with memory complaints found no benefit on memory or biomarkers. The one clean human test of a spermidine supplement didn’t deliver. (hype)
- Diet likely explains much of the signal — Spermidine-rich diets are also rich in whole grains, legumes and vegetables. Some of the "spermidine" benefit is probably just a healthy dietary pattern — and eating those foods is the safe bet. (note)
What we still don’t know
- Does supplemental spermidine do anything? The one dietary-dose RCT was null; higher doses are untested for efficacy and long-term safety in humans.
- Is it the spermidine or the food? Because spermidine-rich foods are healthy anyway, isolating the compound’s own effect is hard.
